I'm getting the exact same error message after installing DNN 4.4.1.  Trying to upgrade from 4.0.3.  Did you have any success figuring out how to fix this issue?

Have not figured it out yet, but I suspect my ISP does not have my permissions set correctly.  I asked them to give ASPNET read, write, modify perms to the virtual root and they said they had but really had not.  I am waiting for them to complete this so I can upload everything again and try it.  You may want to double-check perms on the virtual root as well.  My ISP is kind of slow with support so I have to keep nagging.

Not yet.  I was hoping that my ISP had something set wrong.  I had deleted all my files and db, uploaded and recreated them several times with the same error.  I am a little disappointed in DotNetNuke.  This is ridiculous.
