I tried to install DNN4.5.1 install and source, none of them success, shows following error:

Server Application Unavailable

The web application you are attempting to access on this web server is currently unavailable.  Please hit the "Refresh" button in your web browser to retry your request.

Administrator Note: An error message detailing the cause of this specific request failure can be found in the application event log of the web server. Please review this log entry to discover what caused this error to occur.

I checked application event log, shows:

Failed to execute the request because the ASP.NET process identity does not have read permissions to the global assembly cache. Error: 0x80070005 Access is denied.

But  aspnet has the full control right in my web project folder.

I searched google, someone suggest to modify the machine.config, but I do not want to modify machine.config, since I need to deploy my DNN4.5.1 project to QA and Production server later.

any suggestion?

ASPNET 2.0 uses NETWORK SERVICE instead of ASPNET as the user account by default. Try giving that account permissions on your project directory.

I assigned full control to both NETWORK SERVICE  and ASPNET accounts, but still same error.

Failed to execute the request because the ASP.NET process identity does not have read permissions to the global assembly cache. Error: 0x80070005 Access is denied.

I figured it out:

1) Open IIS
2) Select the Folder
3) Right Button Click on the Folder
4) Click Permission Wizard 
5) Choose Public WebSite
6) All should be ok

it works now.
