I can swear... i leave my site up and running yesterday night at about 24:00.
This morning I try to connect and obtain firt the .NET friendly exception page, then the dotnetnuke page:

try to restart the application (make a minor change in web.config and save it) or restart the web server. If this doesn' t help I assume an issue with the skin of your site, that might show up after the latest security patches from Microsoft. In this case contact the skin developer.

I've already tried the web.config change but without results.
In the second case, this means that changing the skin solve the problem? How can I change the skin acting directly on DB ?

Do you know which are the security pathes that cause the problem?

you can change the skin and container settings directly in the database by removing the entry in the portals table. HTH

Ok. I'll try.
May you give me more information about the security patches that cause the problem?
Otherwise I don't know what to say to Tyler Fawcett (the author of Tranquility2007 skin).
