Would anyone be able to advise on the number of Users they are able to support on
with a DNN site connected to a SQL Server 2005 Express database?

I know there are a lot of other variables but any experience of running against SQL Server Express 2005 would
be very helpful.

This is a hard thing to directly answer as It really depends, but have a look at this, it outlines the "limitations" that are imposed with SQL Server 2005 Express http://searchsqlserver.techtarget.com/tip/1,289483,sid87_gci1082707,00.html  These limitations are around memory and other usage.

If you want to try and get a baseline you could setup a local environment and do some load testing with the free load testing tool from Microsoft.
