I'm a little unclear how this works. Am I correct in assuming that I can create a role which matches the name of one on AD, and they will then map over to that role?

Exactly. The only hitch is that it has to be the Pre-Windows 2000 group name if they're different. I.E. In the AD the group name is Business Staff but if you look at the properties of the group the name is BUSSTAFF then you'd have to use BUSSTAFF as the role name in DNN.
