I am not sure if I will be able to achieve this but I will put it here

I have a website (www.mysite.com) with DNN

now, for every user, I want them to able to login with

user1.mysite.com

user2.mysite.com

a) I am not looking at subdomain configuration in IIS or host headers

b) Not looking at child/parent portals

May be I can get wildcard DNS mapped to my site, like *.mysite.com to IP

Is it possible, that I could portal alias? or a custom HttpHandler?

I am not 100% sure what you are asking here.  

Are you saying that you want your one site to be accessible by basically every possible domain name combination ever.  Such as whatever.mysite.com.  And all of these sub domains point to the exact same site?

Can it be done or we need a portal alias created for every combination

I would think that you would have to do this on the server side as in IIS.

well..if I have to do on the server side, then It will not be dynamic.

hmm...I guess, I will have to play around to give the user an illusion of sub-domains.
