Sorry if this is an overtly obvious question, but do .htaccess files work with a DNN portal? I just upload in with the file manager? Any help is appreciated, thanks.

Unless I am missing something, .htaccess files don't work with IIS.  Are you trying to run DotNetNuke under Apache?  DotNetNuke is designed and tested to run under IIS on windows.

Oh ok.

Well basically I've created a completely remodeled version of a website hosted on a local server.  I'm now ready to move that online to the web host. I'm keeping the same domain name as previously used but many of the individual page names will be changed. I was looking for a way to transfer over the PageRank and Google SERP standings to the new pages using 301 redirects. Is there another way to do this with DNN?

If you control the box, you can use ISAPI_rewrite with 301 redirect rules in .htaccess.

Or you can do it on the DNN level with Url Master.
