I'm look for a way to turn on/off the Partial Rendering for my module from code behind? e.g. when I click the off button, it sets enablePartialRendering = off. Is there something like this I can use? Thanks

You can get referrence to ScriptManager from your module code. and use EnablePartialRendering Property

I heard that I can only enable/disable this on init() ? Is that true?

Without trying it I would say thats correct. You should give it a try.  

Take a look at DotNetNuke.Framework.AJAX.WrapUpdatePanelControl() I have used this in the past from the Load() event.
