Also had the same problem, solved it by going to "Control Panel" -> "Administrative Tools" -> "Internet Information Services". I selected the website wich had DNN 5.1.4 installed, clicked "Properties" and on "ASP.NET" tab i changed "ASP.NET version" option from 4.0.30319 to 2.0.50727.

Had the same error on my DotNetNuke 514 but in install version (Not trying to compile in VS2010)
the following worked for me:

-Go to "Control Panel" -> "Administrative tools" -> "Internet Information Services"

-Select your Website, Right click -> "properties" then go to "ASP.NET" TAB and on "Asp.net Version" change to 2.0.50727
And that Should do it.
