To all:

Is there anyway to force the username to be the first letter (of the firstname) and Full Last name after or during registration. Example: John Doe would be JDOE for the username.

 I am using the default build in registration from DNN. I am on Version 5.2.2 +

Not without customizing the forms or using a manual process to assign accounts (closed system).  You might look on Snowcovered for a third party module that would do this.  Besides, how would you "force" John Doe and Jane Doe to both use the same user name?  :)
