I have DNN 4.x running on IIS 6.0 w. SQL Server Express.

If I havent opened the website for a while, it takes a long time to open the home page (30 secs - 1 min). Is there any guideline to which cacheing parameters/settings either in IIS, SQL Server or DNN that I can tweak to improve performance?

initial load time of ASP.Net applications is increased due to necessary compilation. This is repeated everytime, when the web site is restarted or has not been visited for a certain period (AFAIR 20 min) and gets recycled. To prevent this, you can use any service to call you site about every 15'.
