I would like to see an error message with file name and line number. Normally, when I work on regular site, setting customErrors mode="Off" works but not in DNN. Also, I would greatly appreciate if you would point me to some document on how to setup debug in vs2005 and local IIS.

There is one more question, and I would hold my breath to wait for your response. I am modifying the register.ascx.vb and register.ascx file to add a couple of texbox controls and a submit button to authenticate a user against an existing database before allowing him to register to DNN portal. Everything seems to work nicely until I login and try to unregister from the modify user account screen, I got the below error. Will some one please tell me where I should look at and how I make dnn display a friendlier error message.

Ensure that <compilation debug="true" /> in web.config.
Are you running 4.0.2? If yes, install 4.0.3- it has fixes for caching, that possibly can help.
You also can look in Admin/Log Viewer to see detailed errors.
You also can improve error reporting for developer -see Show exception details during development in DotNetNuke
