In an older version of DNN 4.something I installed to a virtual Directory and then created a new site with the same name, pointed it to the same physical folder as the virtual directory and then deleted the the virtual directory. DNN then ran on its own site.
 
I have tried doing this with 5.5 but haven't been able to get it to work.

Is such a change recommended and if so how is it done?

This is possible, you need to be sure to change the Portal Alias for the new URL

Hi Chris

Thats what I thought but it still didn't work. In the end I have fixed it by logging into the virtual directory version of DNN and adding a second alias in there. Then I stopped the virtual directory site and created a new site pointing to the relevant folder with the new alias. seems fine now
