When I try to edit \App_GlobalResources\GlobalResources.resx through language editor. I get this error...
A File Operation Failed. Please Check Resource File Permissions So Files Can Be Edited.

I checked permissions on that file and everyone have full rights.

What can be the problem here?

Is this in production or a test environment? If in test you might try resetting the application pool in IIS to see if for some reason the file is locked.

did you also make sure, the file is neither write protected nor blocked?

I tried to restart Application pool, but it is the same.

File isn't write protected and how do you mean if file is blocked?

if files are downloaded from the web, they are blocked in recent OS versions. depending how you unzipped the files, they might get blocked, see lower part of first tab of file properties.
