I added "UpdatePanel" to "view" ASCX of my Module. Everything is working fine (I have cascading drop-down lists) only for SuperUser in "Edit" mode. in "View" Mode entire page is refreshed, same for all other Users (with "View" & "Edit" permisisons). I am using DotNetNuke long time, but the last versions are not friendly for development. If AJAX UpdatePanel cannot work inside ASCX control, it is not good. "Partial Rendering" is enabled for Control in "Edit Extension" .

please check the module cache settings - if your module is using caching then the cached copy of the module will be served up (and hence your ajax will not work) - caching is disabled in edit mode which is why it feels like this may be the issue - check this by editing the module control via host->extensions (or checking your dnn manifest file)

Yes, you are right

It was "Cache" option in standard "dnn" manifest file

Without module caching "Update Panel" is working as should be
