I have DNN set up on a VPS, with one portal working fine.

I have created and configured a second portal through the hosts menu when I was logged into the first portal. The domain name for the new website hasn't yet been delegates, but I can access the site via a windows HOSTS file which circumvents the DNS lookup and sends the request directly to my VPS.

Unfortunately, when I go to that URL, I get an error. I'm checking errors through failed request tracking, details follow:

Authentication: anonymous
Trigger status: 500
Final status: 200
Event: module set response error status
Module name: WindowsAuthentication
Notification: 2
HttpStatus: 500
Notification: Authenticate_request
ErrorCode: The operation completed successfully. (0x0)

What am I missing? Does DNN do a check on the back end to make sure the domain is delegated prior to serving content? Is there any other way to preview a site prior to domain delegation?

make sure, you did not enable windows authentication for the web site, unless you need it.

Hi Sebastian,
the Active Directory Authentication module hasn't been installed.

Any other ideas?

I have gone into IIS and into the site, and saw that Windows Authentication was enabled. I have disabled it, and restarted that particular website, but I am still getting the same error. Do I need to restart IIS entirely?

Restarting the app pool should be sufficient, are you sure you are getting same error?
