I'm looking to implement an alpha listing and department listing and I am not sure how to approach this.  The alpha listing should list the alphabet A-Z in a container to the left side of the directory where users can click "A" and it will automatically search for all users with display name starting with "A".  Same with the department list, but this should be put into a dropdown menu if possible.

You should consider a module like xMod which gives you a lot of flexibility in this kind of scenario. You can find it on the dnnstore.
