When attempting to add a portal to an existing site on DNN 7 I receive the following error: Access to the path 'E:\Websites\HSD_External\Portals\_default\Default Website.template.en-US.resx' is denied.

I am logged in as a superuser. My account has full access rights to the default template file.

Any ideas?

Is it DNN 7.0.0 or 7.0.1 or 7.0.2 ... 7.1.0, 7.1.1 or 7.1.2 ? 

Check physical permissions on the dnn root be sure proper permissions are inherited to all subfolders and files.  Check if the file exists on the physical Folder.

I am on DNN version 7.0.5, the template file exists and access rights are inherited for all subfolders and files

Please check the eventlog for error Messages.   Check if the database user on advanced Settings of SqlServer/databasename/security/users/username (Standard is dbo) is mapped with the LoginName you can found on SqlServer/security/Logins/loginName (should be the username you are using on the Connection strings).
