I'm facing to the same problem.

I have a dev instance which is a copy of a pre-prod instance. I upgraded it to 7.2.1 and I've encountered no problem. I made the same upgrade on the pre-prod server and its File Manager doesn't work anymore.

I'm wondering if the .Net 4.5 isn't needed for those features. I noticed that other actions like 'Tools > Clear cache' doesn't work too.

Is .Net 4.5 installed on your webserver? Did you managed to solve this problem?

Hi! I had the same issues on upgraded sites.
I don't know if this solve you problem:

I had some customized modules (inside my web.config->httpmodules) and after various tests i tried to comment that lines:

and then filemanager started to work correctly.

If this is the case i reccomend you (if you can) to debug your custom modules or to try a workaround to correct handlers that could conflict with "DigitalAssets/API/" call .
