Asp.net has a rich set of controls and library, but I see some of developers uses CMS like DNN. I prefer to create a web site from the scratch rather than CMS because if I use CMS, I must learn new topics(for example for DNN architecture).

Some developers says that DNN has a lot of free modules which help create a web site easier and faster. But i think that features are available also in ASP.Net Web Form.

So my question is what i lost when i use ASP.Net Web Form instead of DotNetNuke. What they lost that do not use ASP.Net Web Form?

Creating a website based on DNN (or a similar CMS) allows you to concentrate on function instead of dealing with security (user accounts, permissions, roles), url rewriting, configuration, skinning, caching, ...
Many requirements might be fulfilled by using existing modules and providers.
You may setup a new site immediately, install modules and skins, add pages and modules, even just to mockup UI and present it to your client.
While creating your own modules, you may rely on DNN API and controls, e.g. creating your own settings page just requires you to create an ascx page, a load and store method and registering it via UI.
Create a local install (using install package) of DNN Platform, get DNN module templates from NUGet and start your first module - and you'll see the benefits, which make it worth learning the additional bits of DNN API.

Thank you, But I have still problem with your definition. To clarify this assume I want to create an 'Real State Website' which user can offer house, property, shop-store, and so on. So what functionality DNN provide for me to create this site easily and more importantly faster. Which modules came to help me specially?

DNN will take care of skinning, user and roles management, url rewrite.
The admin might send out Newsletters to all registered users.
You should have a look at property agent from www.ventrian.com
Other interesting modules might be NewsArticle, Forum and one of the store modules.
You may setup user groups and activity stream as well.

As I know asp.net support User and role management and I think user management in dotnetnuke is based on asp.net user management. For skinning master page is equivalent in asp.net. For url rewriting there is some third party module like  http://urlrewriter.net/.
