If you want to install a new web site, the "Install Package" is the option to take. For upgrading an existing web site, you should use the "Upgrade Package".

If you are completely new in administering IIS and SQL Server, you should try to use MS WebMatrix.

Anyway, I would recommend to set up DNN on in a testing environment, "play" around to find out how it works and then go live with a production site.

I agree with Eric, DNN is a great tool for setting up an intranet information system.
