Thanks. Just to be sure by, 'If you have a default domain set' do you mean a populated AD_DefaultDomain in PortalSettings for the (single) portal? If that is the case then it is populated. I had hoped that by removing the domain from each Username in Users then the login mechanism would automatically use the AD_DefaultDomain but that is not happening and login is denied unless domain\username is used.
Can you suggest anything I should check either on DNN or AD side?
