OK, I did a little more digging but with no luck. Here's what I tried (in all cases I made sure I changed the identity used for the app pool in use and set Modify permissions on the \dotnetnuke folder for that account)..

. Switched the app pool to NETWORK SERVICE
. Manually added a new app pool, switched the DotNetNuke app to use that app pool (permissions set to "iis apppool\"
. Tried "iis apppool\defaultapppool"

None of these worked.

To reiterate, this OS installation is as straightforward as I can make it (fresh install, ASP.Net role added).

I am totally blocked on this right now.

you need to run your web site in an app pool, configured for ASP.Net 4 in integrated mode.
In IIS App Pool advanced settings you may specify a user account (identity) for the ASP.Net process. Easiest is using Network Service, if sites from different owners are hosted, you may prefer using app pool identities (IIS AppPool\[AppPoolName]).
This user should be granted modify permission for the DNN directory including all subdirectories prior to running the install process.
If you want to use attached database with integrated security in your sql server connection, you need to create a sql server account for this identity and grant dbowner permission for the newly created database to use.

Thanks for the response, Sebastian. I think I have everything you suggest. Using NETWORK SERVICE as an example of one identity I tried, here's what I have done...

. In IIS -> Application Pools edited the identify of the app pool to be "NetworkService"

. Made sure that the DotNetNuke application (under Default Web Site) uses that app pool (through Advanced Settings -> Identity)

. Set file permissions on the c:\inetpub\wwwroot\dotnetnuke folder (and all subfolders and files) to Modify for the NETWORK SERVICE account

. Restarted IIS (just for the heck of it)

No luck again - same error (as if it can't reach the web.config file).

I should add that the app identity is indeed using .Net 4 and in integrated mode. Also, I manually checked that the NetWork Service account has access to web.config from when I set the permission on the DotNetNuke folder and it does (Modify permissions).

did you make sure, Default.aspx is configured as Default document for the IIS Website?
