OK, I made some really good progress to get to the specific error by allowing IIS run the app in debug mode with IIS providing detailed errors. Here is the specific error with running the 4.5.5 DNN application in a ASP.NET 2.0 application pool in Integrated mode with Networkservice as Identity and load user profile as True. I also set the enable 32-bit applications equal to true for this app pool. Here is the specific error I am getting related to the application path: 

Server Error in '/' Application.

Request is not available in this context

Description: An unhandled exception occurred during the execution of the current web request. Please review the stack trace for more information about the error and where it originated in the code.
