It appears that you are using Human Friendly URL's and have the free iFinity Friendly URL Provider as the default URL provider.

Changing from IIS 6 to 7 as well as moving from ASP.Net 2.0 to 3.5 SP1 will make significant number of changes to your site's web.config and may be the cause of this issue. That would be particularly likely cause if you are using an older version of the iFinity Friendly URL Provider. I would suggest you check their website for support and the availability of an upgrade. I do recall that earlier versions of the free iFinity Friendly URL Provider did have some 404 issues when creating new pages in the early 5.1.x versions of DotNetNuke.
