OK. I see what you're trying to do. If your .aspx page inherits from System.Web.UI.Page rather than from DotNetNuke.Framework.PageBase, you'll need to add a reference to the DotNetNuke.dll and and an Imports statement for DotNetNuke to be able to access ModuleSettings.  You'll also need to pass the ModuleId in the querystring of the url calling this page.  You should then be able to do something like (untested):

If Not Request.QueryString("mid") Is Nothing AndAlso Request.QueryString("mid").Length > 0 Then
     Dim ModuleId As Integer = Integer.Parse(Request.QueryString("mid"))
     Dim mc As New Entities.Modules.ModuleController
     Dim ModuleSettings As System.Collections.Hashtable = mc.GetModuleSettings(ModuleId)
     Dim MySetting As String = CType(ModuleSettings("MySetting"), String)
'Use MySetting as desired . . .
End If

I would also suggest that rather than using an .aspx page for streaming content like this you consider creating a generic HttpHandler (.ashx file).  It is much lighter weight than a full-blown page resulting in a  quicker response.  I use a similar techique for dynamic image creation and streaming - particularly for variable size thumbnails. In the handler's ProcessRequest method you can obtain the Request and Response objects from the HttpContext that is passed into the method and then use code similar to the above.  Other DNN related settings values may be obtained as well - look into the DotNetNuke.Entities.Portals.PortalSettings and DotNetNuke.Common classes in particular.
