By the way, one thing I have done different on the site (and not on the test) is that I installed the Wiki as part of the DNN installation.  I then uninstalled the Wiki.  To reinstall it I had to down load the latest version from codeplex (same version 04.05.00).

Some good news - I have made a bit of progress. 

I am using the iFinity Url Master module and had the 'Change all Urls to lower case' setting ticked.  Unticking this and I can now save the Wiki pages.  I did like the lidea of all lower case Urls, but I guess I will have to do without.

This still did not resolve the issue of results not appearing in Search.  Still need to get this fixed. Then I'll be happy again.

On the search issue, I have tried to procedure suggested by Sebastian in the following post without success:

Except for Pingle, this has been a bit of a conversation with myself.  Anyhow, I have given up on the DNN search [at least for a while] and gone back to using Google Search.   Now I have the Wiki pages available on site search.

Just as a note, I have checked out Canam Wiki and despite having more features, I do prefer the core Wiki.  Using the same HTML editor is the big thing - no markup, same editor as the other modules, etc.  The core Wiki is also not that far behind on features - a good development push and it would be there.
