Thanks Scott. I already rolled back my site. I will create another test instance and test it then. I have always installed DNN the same way in my server and this was no different until now I never had this problem. But if no one else is having problem obviously there is something wrong in my Server environment. Thanks for the quick response by both MVP's.

 the problem is to identify the source of the access error - it might be missing permission of the asp.net user to delete the file or the file might be occupied by a different process - e.g. ASP.Net scanning for changed files which require a recompile (unfortunately, portals subtree is not excluded from this scan, due to skins and containers as well as module files may reside inside.

I am experiencing this exact same issue, my event log is being flooded by errors from the PurgeModuleCache scheduled event.  This is happening with three different production websites that I upgraded to v5.2.1 a few days ago.  I only noticed because my database blew up beyond its size limit for one of the sites and started throwing errors itself.

Any suggestions on how to pursue this?

One thing of note:  this happened exactly when I upgraded to 5.2.1, which included changes to the web.config to add the following providers -

    <moduleCaching defaultProvider="FileModuleCachingProvider">
      <providers>
        <clear />
        <add name="FileModuleCachingProvider" type="DotNetNuke.Services.ModuleCache.FileProvider, DotNetNuke.Provider.ModuleCaching.FileProvider" providerPath="~\Providers\ModuleCachingProviders\FileModuleCachingProvider\" />
        <add name="MemoryModuleCachingProvider" type="DotNetNuke.Services.ModuleCache.MemoryProvider, DotNetNuke.Provider.ModuleCaching.MemoryProvider" providerPath="~\Providers\ModuleCachingProviders\MemoryModuleCachingProvider\" />
      </providers>
    </moduleCaching>
    <outputCaching defaultProvider="FileOutputCachingProvider">
      <providers>
        <clear />
      </providers>
    </outputCaching>

for testing switch to memory caching provider - do the errors go away?
