I tried turn off compression and other settings proposed in this thread. But no success. The solution for me was to delete all the temp-files of the browser (found tipp here: http://www.ventrian.com/Support/ProductForums/tabid/118/forumid/16/postid/17286/view/topic/Default.aspx). Now the portal is running again.

This appears to have worked for me also, first I made all the setting changes in DNN then I closed my browser, restarted my browser, cleared Temporary Internet Files, closed and restarted my browser, logged into DNN used the "clear cache" link under performance settings and then restarted the application from the link on the host settings page.
