I just installed 4.4.1 While everything works well yesterday, I got a "An unhandled error has occurred" error on the first visit today. Then it works well after the second visit. The log says: "Application Started" at the time of first visit.

What I think is that, the app stopped running after a certain time period of inactivity. Is there any way to solve this problem?

I have disabled the scheduler.

ps. I installed the DotNetNuke 4.4.1 install package in my shared account on HostingFest.

The fact that ASP.NET's worker process stops is a "feature" of ASP.NET.  There are a few things you can do to limit the frequency of ASP.NET process stops.  I use a keep alive service from http://www.host-tracker.com (Free).  They request a specific page on my site every __ minutes which keeps my site alive.  This works fairly well, there are a few other sites out there to do this type of thing as well.  KeepAliveForever.com is another one that comes to mind.
