I just completed a 500 person migration from another system to DNN.  I didn't want to do any direct manipulation of the DNN tables and found a simple import file.  I extended it include roles and do some more reporting.  You can see how I did it at: http://www.clickandcreate.us/People/EricShafer/tabid/161/EntryID/12/Default.aspx
