Pretty clean IIS 7/Vista install. 

Downloaded DNN 4.5.1_Install

Started SQL 2005

browsed to my application on the localhost and *BOOM*

Server Error in '/dnn' Application.

I'm running into this exact same problem, so if anybody has a solution, that would be great.  In try to find out what's going on.

Having the same problem too.  Found a workaround.

Running Vista / IIS7 / Sql Server 2005

Downloaded 4.5.2 Install package, and followed the steps in the install doc.

Browsed to the site and got the exact same error as EchoBinary above.

In IIS for the dotnetnuke application that i just created, in the advanced properties, i changed the application pool to use Classic .NET AppPool (which has a Classic Managed Pipeline Mode) instead of DefaultAppPool (which has an Integrated Managed Pipeline Mode) and it started working.

Not sure why exactly.

Check whether the DNN application is configured for the the dot net framework 2.0 in the IIS.
