Is your site running on D drive on local and remote? The DotNetNuke.config file is located in the config directory and I have been getting a lot of these errors as well. By just copying the config file to the root directory from the config directory the site should run just fine. If your looking for the reason this is happening - I don't know but you can ask that question in a different post.

Good luck,
