I Have the same Question post at http://support.dotnetnuke.com/issue/ViewIssue.aspx?ID=5966 and here i get the answere:

Clear cache, restart app pool.

Thats working.

Changed webconfig (changed trust level twice )  the host tab got visible. Odd..

it's very simple: Clear cache, restart app pool.
