I do not understand why I am getting this error....

"A critical error has occurred.
Request for the permission of type 'System.Security.Permissions.FileIOPermission, mscorlib, Version=2.0.0.0, Culture=neutral, PublicKeyToken=b77a5c561934e089' failed."

Does anyone have a clue how to fix this?

Me too!  It seemed to be intermittent and I've been searching and haven't really found the answer but I did try this....

I'm just running this on my own development machine to test it so it's not on the Internet.  I'm running Windows XP Pro with IIS 5.1.  I never set up a domain and I can't find how to give NTFS access to ASPNET.  All I can find is a way to share the folders.  Problem is that sharing a folder does not seem to share the subfolders. When I went in to my DNN folders and shared the subfolders (and their subfolders . . . .) this problem seems to have gone away.  I'm just a newbie poking these things with a stick to get it to work.  I'd appreciate it if someone who knows what they're doing could advise us both.

I am also recieving this error and can not get past an initial login because of it. Then it seems after a few navigational clicks I can not bring up the front page at all. The 4.5.5 install went smooth with no problems or errors. The remote host I am working with is not using SQL Server 2005 Express but I did change the appropriate connection strings as the documentation states. And the file permissions are set correctly via a applet from the host. I'm at a loss of what to do next. Any help will be apreciated. Thanks.

Make sure, your virtual user "ASPNET" or "Network Service" has read, write, delete and create folder permission for the installation directory and all subfolders.
