If you refer to the DNN Blue skin, you can see the code they have used within a table cell to specify a content area. - All you need to do is copy this method & build a table so that it has the content areas you require. If you don't add any content, DNN will automatically shrink that table cell so it will not affect the layout. - So you can just use one skin to create the layout you require.

As a quick tip, the line of code you are looking for to add to your new table cell is:

 <TD class="toppane" colspan="3" id="TopPane" runat="server" valign="top" align="center">

So you just rename the id to another id that is not currently used and this will create the new content area for you. - If you need to specify a special css class setting for that content area, then also rename the class to your new id.
