It seems IIS is not setup to take your domain.  Either that or your firewall is blocking it from the outside world.

Ok I am getting somewhere now maybe. I am able to access the default.htm page by going to www.mydomain.com but I still cannot access my www.mydomain.com/DotNetNuke by that direct link or by just the www.mydomain.com

I can now access my site at www.mydomain.com/dotnetnuke but I still can't access it at www.mydomain.com which is what I wanted to do.

This is because your IIS virtual server is pointed to the wwwroot folder.  You will have to go into your IIS properties for that virtual server and set its default directory to the wwwroot\DotNetNuke one.
