Just downloaded the DotNetNuke_04.08.01_Source.zip version and tried to build. Got this error at :

 

Private Sub Application_Start(ByVal sender As Object, ByVal e As EventArgs)If Config.GetSetting("ServerName") = "" Then  ***********************Here
ServerName = Server.MachineName
Else
ServerName = Config.GetSetting("ServerName")   ***********************Here

End If
End Sub

I am getting the same error on DNN 4.09 source locally on Vista. 
Trying to figure it out now.

this is usually hinting to an issue in IIS web site / virtual folder configuration. Do not nest DNN within another ASP.Net application and make sure, not to have another web.config file within any directory of your DNN installation.
