Existing users can login to the site, but new users aren't added. In the logging I see much errors of the same type:

The funny thing is that when I check the settings there is also an error.

Is this an upgrade from a pre DNN 4.6.0 install? It looks like either you've uncommented the wrong line in the web.config (though if memory serves that'll throw an error just trying to load the site) or the old authentication.dll is still in your .bin directory.

On the settings page, does that error prevent the dropdownlist boxes from populating? If so then I think I know what it is. Open the database manually and open the ModuleSettings table. Find the SettingName AD_ProviderTypeName and make sure that it has the SettingValue of DotNetNuke.Authentication.ActiveDirectory.ADSI.ADSIProvider, DotNetNuke.Authentication.ActiveDirectory
