It sounds like you are going from SQL Express to SQL Server. You will need to update your web.config file to point to the new database. SQL Express allows user instances, versus SQL server which will require you to supply credentials for an account that can authenticate to the database.  It's usually best to create a log in within the SQL security folder and give that log in access to that single database. Remember to update the connectionStrings section as well as the appSettings section of the web.config. Once you do this, the site should kick right in.
