Yikes!

Assuming you have a vanilla install with no DNN-UI changes (which sounds unlikely) you will be safe unzipping a new copy of this file (or the entire directory, if you're feeling lucky) from the DNN install package.

Try to force a reset (modify your web.config file).  I've had this on my hosting provider a couple of times and it ACTUALLY has to do with corrupt files in the temporary asp.net folder.  After a couple app-domain resets it corrects itself and as of yet they haven't determined what it really causing it.  Pretty strange though.
