DotNetNuke should work in Medium Trust environment, but it doesn’t on default installation (checked on 4.9.0, 4.9.3, 5.0.1).
There are few symptoms : redirect loop error, not working some modules which use AJAX (ex. feedback module), error page on DNN 5.0 and so on.

I've figured how to fix this issue (at least on IIS7) and want to share with you, see my blog post:

Alternatively simply install MS AJAX for .NET framework 2 on your server and it will work out of the box. The workaround in the URL provided in the previous post will not work with some third party modules that don't like .NET 3.5 SP1. However .NET 3.5 SP1 is the only way forward and those module developers should be making them friendly for .NET 3.5 SP1 as soon as possible.
