Can someone not only tell me how to fix this but what these requirePermission and their paths point to and do.

I'm using asp.net 2, nuke 4, and win 2000 sql server

I had a similar problem with DNN4.03 installing from the starter kit. Devstudio had asked to upgrade the site to asp.net 2.0 but seems this hadn't occured. Simply changing the asp.net settings for the site in IIS got rid of this error.

You may have the right version of ASP installed, but you may still find your IIS site is set to use the old version.  In IIS snap in, choose the appropriate web site, right click > properties.  Choose the ASP.NET and check the version in the drop down, change to v2 if required.  It just fixed it for me!
