Yes it will. Change your IIS application pool to ASP.Net 4.0 then hit the site with your browser. When the site restarts it should sense the change and automatically reconfigure the web.config to run under 4.0.

You may, however, find a few core and third party modules which will not be compatible with the change and in particular the much stricter parsing of markup, changes in rendering, etc. The DNN Gallery module is one of those. Upgrade the module first to the 4.03.01 release candidate version available at http://dnngallery.codeplex.com/releas....

To avoid issues such as that, you may wish to first clone your existing site to a localhost install and try the ASP.Net 4.0 changeover on the cloned site, work out any problems, then try changing your production site. Also, be sure to make a filesystem and database backup of the production site before any major change such as this.
