Something very strange just started occurring on our DNN 5.x page. Simply put, when logged in as an Admin, the Admin is no longer able to manage the roles for the site users. What's unusual is that none of our other DNN-based pages are like this, so I know for a fact that something is screwy with this particular. 

Just for clarification, this is the message we're seeing when we try to manage the security roles for a user on the site:

Access Denied: Either you are not currently logged in, or you do not have access to this content.

Would anyone happen to know a fix for this?

Have you checked the role permissions for that Admin User?  Should be the first thing I want do and check if there is added a expired day for the added Administrators role.

User "admin" has the following roles:

- Administrators
- Registered Users
- Subscribers
- MyPages Managers (for one of our modules)

Have the admin still access to the other admin pages ?  Works he on the Edit page mode, not on View or design (On the Top and left site)?

 Yep, the Admin can still access all of the usual pages like normal. The only page the Admin cannot access is the "Manage Roles" for a given user. I double checked and the page is set to "Edit" mode.
