Rob, Of course I understand that it should be working properly. It was just my 2c in case it is urgent for you to make sure you’re aware of this trivial hack.
Anyways, after many upgrades from 4.9+ to 5+, with quite a few that I had to re upgrade again and again, I concluded that this kind of problems is a compatibility issue. I am sure that you are aware of that, but I would try removing modules one by one from the installation until you’re left with a bare bone installation.
(I see that even Sebastian is running out of ideas and that’s a good sign to start chopping)
BTW, you said “DNN v4.9.5 to v5.5.0”, did you follow one of the upgrade paths like Will Strohl’s  http://www.willstrohl.com/Blog/EntryId/102/Suggested-DotNetNuke-Site-Upgrade-Path-s  ?
What was the version of the first install of this DNN?
Did you upgrade all the modules you could to the last versions?
If it started from, say, v3.3, it is probably littered with files and definitions that you don’t want.
Just as an anecdote I can tell you that one of Inventua older modules was a show stopper for an upgrade I had lately.

This portal started out as an early 4.0 site.
Yes, I have been very careful to investigate all of the modules to ensure I have the latest version that should be DNN 5.5 compatible. I even tested most third party modules on my other working DCC v5.5 sites to be sure. This is one reason I have waited so long to attempt this upgrade. not smooth sailing.

I could not upgrade this from DNN 4.9.5 to DNN 5.5.0 directly as I found a compatibility problem with the Events module. Went from 4.9.5 to 5.2.3, upgraded Events to latest version, then I tried to upgrade to 5.5.0. Still problems with a sql data provider (5.3.0).

I then looked thoroughly and  found lots of old sql tables, sprocs, dlls left hanging around from over the years. cleaned all that up, and the upgrade from DNN 5.2.3 to 5.5.0 then completed successfully, except for the two problems documented above.

There is no real option to uninstall modules in a production environment if they have a lot of data associated with them. However I could install a couple of additional modules on a test 5.5.0 site to see it that breaks anything. But I'm more inclinde to think there is something else at play here since the vbc.exe compiler is being called during this operation.

I would bet if more users actually tried to edit languages as I am trying, I bet a lot more complaints would pop up. I only have the English language, but I customized some text in certain modules, and even what is sent out during a registration. My experience says very few admins even know about what they can do with the language editor in this reqard.

I did not mean in any way that you'll do it on the production site. It was obvious to me  that you do all the research on a duplication site.

if you are using a single language only, preferred language might not properly set for your portal. did you try to re-save preferred language and site settings?

While watching the w3wp.exe process for the problem site (they all run under different unique user accounts and application pools) I noticed the following. When I click on one of the language edit icons, the vbc.exe process is spawned as a subprocess of the w3wp.exe process, terminates and the error in DNN comes up. (the full path is c:\windows\microsoft.net\framework\v3.5\vbc.exe)

Vbc.exe is the vb compiler, which is called to compile the web site, which gets cached afterwards. it looks like a compile issue, is there anything logged inside the Windows application log?
