I just get following error message:
Index #: 0
Source: .Net SqlClient Data Provider
Class: 14
Number: 15382

I have the IIS AppPool\DefaultAppPool and Network Service full access to the directory and db file. Nothing else is open when I try start the site.  What else can I check.

did you make sure, SQL server allows remote access and TCP/IP is configured for SQL Server and client?

This is on development machine and not being accessed remotely. Would that still be an issue?

you don't need remote connection to access it from same machine, but TCP/IP

I ran into the same problem.  Very frustrating.  Solved the issue by going to:

http://support.microsoft.com/kb/2002980

Followed  these instructions and the database issue was resolved.
