Usually a simple change of the ASP.Net version under which the AppPool is running will allow DNN versions 5.04.04 and later to make the correct changes to web.config for ASP.Net 4.0.

Could you post the details of the error that you received with the MAC Viewstate? When running under ASP.Net 4.0, a change is made to web.config to add the attribute/value pair requestValidationMode="2.0" to the <httpRuntime . . . /> node. Did you force a restart of the IIS application after the error to see if it re-occured?

The only other changes made to the web.config are related to binding redirects for the System.Web.Extensions and System.Web.Extensions.Design assemblies. As for storing information about the .Net framework version in the database, as far as I know, this is not done.

One question for you . . . was the ASP.Net 4.0 App_Pool set up for classic or integrated pipeline model?
