I dont know if dnn uses transacations anywhere..  but if so..   you have to be able to do a reverse lookup ..  make sure you can ping the iis server from the db server ...

Great, If it is an IIS issue, then try the following:

1.  Make sure you have timeout=30 in your connection string in the web.config file.  I found it to help in some situations.

2.  Before rebooting the IIS server next time, try and Kill the aspnet_wp.exe process from the task manager and see if this will fix the problem.  If not, there is another process that I don't recall its name exactly "could be W3WP.exe" try and kill it also.

I wonder if this isn't a problem with the Connection Pool.

You might want to read this article, and see if it helps troubleshoot the issue.

I'm running DNN 2.12 still in our environment, but I've noticed a problem that if you can't connect to the SQL Server, you'll get that message about Assembly Version mismatch.  That sticks around until you reset IIS though, at least for us.  That is, DNN caches something and doesn't bother to go back to try again later.  So it's a bad thing.  I haven't looked to fix this and am not sure if it's the case with 3.2.2.
