Is it possible to get DNN to send an automated message to notify specific users when a new document that they have been granted access to is uploaded into the Documents/Admin File Manager module?

the documents module doesn't support that (hopefully when 6.2.0 comes out they will integrate with the notification API to supply functionality such as this), but you could write your own dotnetnuke scheduled task to do so - http://kemmis.info/blog/archive/2008/03/08/how-to-write-a-custom-dotnetnuke-schedulerclient-i.e.-a-scheduled.aspx
